Rhodamine 123 is a fluorescent dye most commonly used in flow cytometry, which can be used as a functional reporter molecule of P-glycoprotein (Pgp). It is part of Rhodamine flurone dyes. [4] This dye is mainly used to evaluate the mitochondrial bioenergy metabolism of living cells. In cancer treatment, the functional measurement of multidrug resistance (MDR) protein is a better prognostic indicator than the expression level of MDR protein. Rh 123 can be used for multi parameter analysis without fluorescence interference, and can be used in combination with common protein labeling dyes, such as PE-Cy5 and AMCA (7-amino-4-methyl Coumarin 3-acetic acid). Recent reports suggest that Rh123 may also be a substrate for multidrug-resistant protein-1 (MRP1).
